They had been practising rock climbing in Montserrat, around 30 miles north-west of Barcelona.
Part of a group of seven climbers, the man died shortly after being transferred to Barcelona’s Vall d’Hebron Hospital.
It has also been confirmed that the woman lost her life in Bellvitge University Hospital.
On Saturday, the Montserrat Mountain Trust announced the immediate preventative closure of the five climbing routes.

Manager Xavier Aparicio said the measure had been taken to ensure police mountain specialists were able to investigate the fall site.
Specialists later revealed there was a high risk of further rockfall.
“The aim is to prevent more accidents,” Aparicio said.
So far the climbers have only been identified by their first names – Marc and Laura.
It is believed the two climbers were both from Barcelona.
After a day’s climbing, the group had been preparing to leave the area at 4.30pm on Saturday when tragedy struck.
Both victims of the rockfall were found unconscious near a parking area called Can Jorba – close to El Bruc on the south side of Montserrat.
Emergency services had rushed to the scene and were able to revive the male climber for a short time before he went into cardiac arrest.
The female climber was rushed to hospital in an air ambulance.
Montserrat has been regarded as a dream destination for rock climbers, attracting enthusiasts from all over the globe.
